A target fragment is excised from the vector or BAC molecule, forming a small loop structure that is inserted into a fixed position within the genome. This insertion is facilitated by the transposase, ensuring stable integration. This technology has gained widespread use in CAR-T cell therapy due to its low cost, high efficiency, and ability to maintain stable gene expression.
